I (probably unfairly) lump those movies in with a genre I really just hate, which is the Saga of the Man-Child. I am so completely over adult men who act like they're still irresponsible college bros -- which is pretty much one of the biggest movie comedy genres in the last decade. But your descriptions make it sound like maybe these aren't that kind of story?
Edit: nevermind, just glanced at the post again and saw "irresponsible men." I had focused more on the part where the guy in Knocked Up gets over his immaturity, and the lead in 40 isn't that irresponsible at all. Sounds like the "comedies to appeal to men as well as women" category is pretty much the Saga of the Man-Child, though.
